Covalent TCR-pMHC Interactions: A New Mechanism for T Cell Antigen Recognition and T cell Activation

Abstract Noncovalent bonds typically mediate interactions between a T cell receptor (TCR) and peptide-major histocompatibility complex (pMHC) ligand. In this study, we describe novel mechanism involving covalent TCR-pMHC interaction through cysteine-cysteine disulfide bond. By introducing cysteine resodues into the TCR CDR3 peptide of known combination, show that bond formation does not require structural rearrangement can still occur even when initial affinity is low. This strongly activates signaling has potential applications in basic immunology to further our fundamental understanding activation/differentiation as well applied clinically for improving efficacy sensitivity adoptive therapies. Our findings shed new light on activation provide additional avenues development modern immunotherapy.
